Ultra Perth takes place at Sir James Mitchell Park and along the South Perth foreshore. The event uses an AIMS-certified road course that organisers describe as flat, with wide footpaths and no road crossings. The route follows the banks of the Swan and Canning Rivers and finishes against the Perth city skyline at the South Perth foreshore.
Key course features named for the route include Booroloo Bridge, Elizabeth Quay and the Narrows crossing. The event offers multiple race formats and starts organised in waves, with new Elite categories added for competitive fields.

A club challenge runs alongside the races: clubs with 25 registered runners receive a free VIP tent at the finish, and the club earning the most points wins a $2,500 donation to a charity of its choice. The organisers provide a spectator guide noting riverfront viewing, coffee spots and parking to help crew and supporters find prime locations along the looped course.
Practical information available for participants includes a race manual, details on check in and transition stations, shuttle buses and cut offs.
