The route covers a roughly 208 mile course that begins at the Pisgah Ranger Station Visitor Center just outside Brevard and finishes at the Nantahala Outdoor Center in Bryson City.

The course crosses parts of five Western North Carolina counties and travels through Pisgah National Forest, Nantahala National Forest, and sections of the Blue Ridge Parkway, including two parkway exchanges. Teams split the route into 36 legs and may enter with 6, 9, or 12 runners.
